ABSTRACT

Aim To evaluate the application of submandibular gland ultrasound in diagnosing mouse model of Sjogren' s syndrome (SS). Methods SS model was established in C57BL/6 female mice. Hie saliva volume, water intake and body weight of mice were measured on d 0, d 21, d 28 and d 35 after the first immunization respectively, then the index and pathology of submandibular gland were detected on d 35. Meanwhile, ultrasonic examination was performed on the submandibular gland on a Vevo 2100 small animal ultrasound imaging system. The correlations between the ultrasound observations and the clinic manifestations or submandibular gland histology of SS mouse were analyzed. Results The saliva secretion of SS mice began to decrease significantly on d 28 after modeling, the index of submandibular gland increased significantly on d 35, and the typical pathological changes of SS were observ ed in submandibular gland from model mice. On d 21, it was found that a marked area increment of submandibular gland in axial section of SS mouse compared with that of normal mice. More blood flow signals were detected in submandibular gland of SS mice on d 28 and d 35. Hie analysis showed an evident correlation between the results of ultrasonic observation and the conventional detection indexes. Conclusions Ultrasonography of submandibular gland is able to provide early clues and sensitive semi-quantitative indexes for e-valuating the pathological changes of SS mouse model. It is of great value in dynamically observing the process of SS and thus it is a promising approach for pharmacodynamic studies of anti-SS drugs.

ABSTRACT

Objective To evaluate the effect of air temperature on non-accidental mortality (A00-R99) and years of life lost in Wuxi city. Methods Data on daily non-accidental mortality and meteorology index were collected from 2012 to 2017. Distributed lag non-linear model (DLNM) was used to assess the effect of temperature on non-accidental death and YLL and the cumulative effects between cold and hot temperature on non-accidental mortality and years of life lost with different lag days. Results A V-shaped relationship was noticed between temperature and mortality. Cold effects were delayed by 3 days and persisted for 14 days. Hot effects appeared acute and reached the peak at the same day. Low temperature had stronger gross effect than high temperature had. There were differences of temperature effects between different age and gender groups. Conclusions Low and high temperature were associated with elevated mortality risk. Cold effect had lagged effect and persisted for long time, however, hot effects appeared acute and the impact of low temperature was greater.

ABSTRACT

Aim To compare high-content cell imaging system and other methods in detecting cell proliferation, including the traditional thymidine ( 3H-TdR) incorporation method, methyl thiazolyl tetrazolium ( MTT ) method and cell counting kit-8 (CCK-8) method. Methods The fibroblast-like synovial cells (FLS) were used as the study object to observe the sensitivity and stability of FLS proliferation in different methods by using the usual proliferative stimulant tumor necrosis factor-α ( TNF-α) and the known proliferation inhibitor methotrexate at different concentrations. Results The 3H-TdR method and high-content cell imaging could detect a significant inhibitory effect of 1 nmol ·L-1 MTX on FLS cell proliferation, MTT assay and CCK-8 method could detect the significant inhibitory effect of 10 nmol· L-1 MTX on FLS cell proliferation. 3H-TdR method was found to have a large degree of discretization in the data set, with a standard deviation of 32.61% ~61.36% , and the MTT method was 11.9% ~ 17.8% , the CCK-8 method was 17.15% ~32.88% , and the high-content cell imaging system method was 12.66% ~26.54%. Conclusion The method of high-content cell imaging system is more accurate and stable for detecting cell proliferation.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the contamination state of Legionella in cooling water samples from different places in Wuxi city and reveal the molecular biological characteristics of Legionella strains.</p><p><b>METHODS</b>112 parallel water samples (500 ml each) were collected from 56 sites in Wuxi city during year 2009 - 2010. The samples were used for Legionella test and quantitative culture. The isolated Legionella strains were used for serotyping, pulsed-field gel electrophoresis (PFGE), sequence-based typing (SBT), and intracellular growth were tested.</p><p><b>RESULTS</b>The positive proportion of Legionella was 39. 3% (22/56) among all sampling sites. A total of 29 Legionella strains were isolated, and the serotypes include LP1, LP3, LP5 and LP6. LP1 serotype was the major one with a proportion of 65.5% (19/29). 29 Legionella strains got 17 PFGE types. There were 10 SBT types among 10 Legionella strains with different PFGE types. Comparing to LP1 strain (ATCC 33152), WX2011062 (LP6) and WX2011067 (LP5) had strong intracellular growth ability in mouse peritoneal macrophages J774 cell line (the amount of intracellular bacteria on day 0 after infection were (5.5 +/- 1.32) x 10(5), (3.9 +/- 0.60) x 10(5), (7.8 +/- 0.76) x 10(5) CFU/ml, respectively; the amount of intracellular bacteria on day 3 after infection were (58.3 +/- 1.61) x 10(5), (2700.0 +/- 655.74) x 10(5), (3066.7 +/- 208.17) x 10(5) CFU/ml, respectively).</p><p><b>CONCLUSION</b>The Legionella contamination existed in cooling water samples from different places in Wuxi city. Legionella strains isolated showed high genetic variation. Some Legionella strains had vigorous intracellular growth ability.</p>
